Kneeshaw, Chris Hunter and Stephen Thompson all scored in the first half, as the Quakers led 3-0 at the break.
Kneeshaw then scored twice more, with Graeme Armstrong coming off the bench to get two goals of his own while Lewis Gibbons also scored.
Brian Atkinson took charge of the Quakers tonight, with Martin Gray away on vacation and the side featured a strong line-up which included trialist’s Kneeshaw and Gibbons.
The Bishop Auckland squad featured ex Quakers Mark Bell, Adam Nicholls, David Mitchell and Adam Cocks.
Darlington started brightly and Kneeshaw opened the scoring after a Terry Galbraith corner wasn't properly cleared.
It was two on twenty five minutes when Hunter headed home from another Galbraith corner at the back-post.
Thompson made it three before half-time finishing well first time after great work down the right-wing by Adam Mitchell.
Despite the score-line, Bishop could have scored multiple times themselves in the first period. Pete Jameson in goal was forced into making several one-on-one saves from Stuart Thompson, Lewis Martin and Ryan King.
Veteran Jim Provett came on in goal for Jameson at half-time.
Darlington dominated more in the second-half and two goals in five minutes put the game to bed.
First Kneeshaw latched onto a flick by Thompson and showed great composure to pass into the corner of the net.
Right-back Gibbons then grabbed the fifth, at the second time of asking, as he got in at the back post to tap home.
Armstrong came on for the final thirty minutes, as Darlington made five substitutions in a short space of time.
With quarter of an hour to go Kneeshaw grabbed his hat trick, tapping into an empty net after Mitchell's shot had been saved.
Armstrong got on the score sheet racing clear to finish at the second attempt ,after his first was tipped on to the woodwork by Bishop goalkeeper Scott Pocklington.
In the final few minutes he then grabbed his second, turning the ball home after more good work from Kneeshaw.
Darlington have one pre-season game remaining, away to Spennymoor Town at the Brewery Field on Friday night.
